我有一個做蛋白質磷酸化(信號通路)研究的朋友,他說:WB是條不歸路,WB難,磷酸化蛋白的WB更難!他曾創造過一個實驗室記錄,做了200多次磷酸化蛋白,出結果的只有60幾次,與理論一致的只有40次不到。他,曾崩潰過...
現實情況確實如此,內源性目的蛋白本身在細胞裂解物中表達豐度不高,而其磷酸化修飾的蛋白含量就更低了!即使用高濃度抗體,延長曝光時間,依舊很難獲得理想的效果。大多數市售的WB封閉劑(如牛奶,血清,BSA)雖然可以滿足阻斷膜上的未反應位點,減少檢測過程中非特異性抗體的數量,但它們并不是為了在印跡過程中保護磷蛋白抗原而設計的,本身含有磷酸化蛋白成分,因而并不能為磷酸化WB雪中送炭!
